Aquaporins are small proteins of the cell membrane which allow a selective transport of water through the cell membrane. Some AQP isoforms showed a high level of expression in tumour cells, particularly in very aggressive tumours. New results concerning the part of AQPs in terms of cell migration processes suggest that the expression of AQP in tumours and tumour vessel has an adverse effect on tumour expansion and metastasis. Repression of AQP could be an interesting new approach to design a target tumour therapy in future.
The C-allele of the promoter polymorphism AQP5 -1364A>C is associated with reduced expression of AQP5. It has to be analysed if there is genotype-related association between the functional AQP5 -1364A>C polymorphism and the clinical outcome.
We analysed the genotypes of 182 patients with squamous cell carcinoma of the oro- and hypopharynx. The distribution of the genotypes was 115 AA, 59 AC and 8 CC. The Kaplan-Meier-analysis and the log-rank-test did not show a significant genotype-related difference with regard to the overall survival (log rank p=0.65), the tumour-related survival (log rank p=0.502) and the relapse-free survival (log rank p=0.649).
However, in our patients the -1364A>C (AQP5) genotype did not show a prognostic significance in squamous cell carcinoma of the pharynx.
