Increased transient receptor potential channels in patients with essential hypertension

Alterations of cellular cation influx have frequently been described in essential hypertension. However, the role of non-selective cation channels of the transient receptor potential (TRP) family in hypertension has not been evaluated yet. We investigated TRPC3, TRPC5, TRPC6 in human monocytes from normotensive control subjecs and age- and sex-matched patients with essential hypertension using real-time polymerase chain reaction and in-cell Western assay. Quantitiative TRPC mRNA measurements were normalized to GAPDH, and TRPC channel protein expression was normalized to CD11a as internal reference. Cation influx was measured using the fluorescent dye technique and quantified as fluorescence increase above baseline. The normalized expression of TRPC mRNA was significantly higher in patients with essential hypertension compared to normotensive control subjects (1.492±0.028, n=27; vs 1.515±0.022, n=23; mean±SEM; p<0.05 by Mann-Whitney-Test). Using in-cell Western assay, we observed a significant increase of TRPC3 and TRPC5 channel protein expression in cells from essential hypertensive patients compared to normotensive control subjects (normalized TRPC3 expression, 3.21±0.59 vs. 1.36±0.07; p<0.01; normalized TRPC5 expression, 2.10±0.28 vs. 1.40±0.52; n=31; p<0.01 by Mann-Whitney-Test). Gadolinium influx was significantly higher in patients with essential hypertension compared to normotensive control subjects (48±4% vs. 36±3%; each n=51; p<0.05 by Mann-Whitney-Test).
