Experimental investigation of doxorubicin and irinotecan loaded microspheres for intratumoral treatment of glioma

Objective: Doxorubicin and irinotecan loaded microspheres are specifically designed to and clinically used for trans-arterial chemo-embolisation of tumors. In a rat model it is investigated whether these microspheres may be used for intratumoral glioma therapy.
Methods: Doxorubicin or irinotecan loaded micropsheres (DC BeadsTM, Biocompatibles, Farnham, UK) were stereotactically implanted seven days after stereotactic implantation of BT4ca malignant glioma cells into the brain of BDIX rats (syngenic tumor model). Survival and histomorphological findings were compared between an untreated control group, a group receiving unloaded microspheres and a group receiving doxorubicin or irinotecan loaded micropheres. Size of groups n=10.
Results: After implantation of Doxorubicin Kaplan Meier Analysis showed a doubling of survival time (p<0.05) while the prolongation of survival time with Irinotecan was not significant. Histomorphological investigation depicted marked regressive effects within the tumors with both doxorubicin and irinotecan. In doxorubicin treated animals in addition a hemorrhagic imbibition of the tumor was found.
Conclusions: Intratumoral implantation of Doxorubicin loaded DC BeadsTM decreased the mortality of BT4Ca tumor-bearing rats. This effect could be related histologically to necrosis of the tumors. The experimental findings have clinical relevance, because such microspheres could be readily implanted by stereotaxy either into the tumor or into the tumor resection borders.
