Question:Endometroid adenocarcinoma with squamous differentiation are rare carcinomas of the endometrium. The aim of the examination was to determine possible pathogenetic factors and molecules, which are relevant for therapy of these tumors. We tested eight endometroid adenocarcinoma with squamous differentiation for HPV, steroid receptors and inhibin/activin-subunits.

Methods:Eight endometroid adenocarcinoma with squamous differentiation were immunohistochemically tested with specific monoclonal antibodies for HPV (polyclonal anti-HPV and monoclonal anti-HPV 18), estrogen receptor ER-α and ER-β, progesterone receptor PR-A and PR-B and the inihibin/activin subunits Inhibin-α, -βA, -βB, -βC and -βE.

Results: HPV 18 and the polyclonal HPV antibody was detected in all endometroid adenocarcinoma with squamous differentiation, both in the endometroid (n=7/8) and squamous (n=8/8) parts of the tumor. Both estrogen receptors ER-α and ER-β were not detectable in any tumor in contrast to progesterone receptor PR-A and PR-B, which were detected in about the half of these tumors: PR-A: n= 5/8 and PR-B: n= 2/8. Inhibin-α und -βB were not detected, Inhibin-βA and -βE were expressed in all these carcinoms and inhibin-βC in nearly all tumors (n=7/8).

Summary:The carcinogenesis of endometroid adenocarcinoma with squamous differentiation could be correlated with an HPV 18 infection. Adenosquamous endometrial carcinomas seem not to be controlled by estrogens. The missing expression of inhibin-α subunit suggest a tumorsuppressive function in endometroid adenocarcinomas with squamous differentiation. The missing expression of inhibin-βB subunit, which is probably a marker of differentiation, points out the malignancy of these tumors. The other inhibin subunits Inhibin-βA and βE were expressed in all and Inhibin-βC in almost all of these tumors. It remains to be seen, if it is possible to use these parameters as tumor markers.
