Hypertensive patients show increased oxidative stress and increased cardiovascular morbidity. To evaluate the underlying mechanisms we investigated whether oxidative stress increases the activity of mitogen activated protein kinase kinase 3/6 (MKK 3/6), p38 mitogen activted protein kinase (p39 MAPK), and heat shock protein 27 (HSP 27) in platelets. Platelets were exposed to 20 µmol/L hydrogen peroxide for several time points and MKK 3/6, p38MAPK, and HSP 27 were measured by immunoblotting using specific antibodies. The administration of hydrogen peroxide significantly inreased MKK 3/6, p38 MAPK, and HSP 27. The maximum stimulation of MKK 3/6 could be obtained after 10 minutes, maximum stimulation of p38 MAPK, or HSP 27 could be obtained after 30 minutes. The activation of HSP 27 could be blocked by SB 202190, a specific inhibitor of p38 MAPK. The study indicates that oxidative stress enhances the activty of signal transduction pathways.
