Objective: Glioblastoma multiforme is known as one of the most malignant tumors of the brain. Tumorgrowth arises from the infiltrating zone of the glioblastoma at the border to the normal brain tissue. MTORC1 serves as important regulator of the cell-metabolism. Key mokulators MAP4K3, RagC and VPS34 were poorly understood regarding to glioblastoma in the center and the infiltrating zone.

Methods: Seventy-four samples of thirty-seven patients were collected and separated regarding to the tumor-center and the infiltrating-zone. Samples were than frozen at -80°C. RNA was extracted and through reverse transcriptase a quantitative PCR was performed. Results were equalised against β2-Mikroglobulin. Together with TMA’s and WESTERN-BLOT’s of some samples were analysed. Differences of MAP4K3, RagC and VPS34 were analysed between tumorcenter and infiltrating-zone. Concentration of MAP4K3, RagC and VPS34 were compared to overall-survival of the patients.

Results: MAP4K3 and VPS34 were equally distributed between tumor-center and infiltrating zone (p<0,01). RagC was expressed more in the infiltrating-zone compared to the tumor-center (not significant). RagC was in both locations less expressed compared to MAP4K3 and VPS34. Low expression of MAP4K3 in tumor-center and infiltrating zone were associated to a longer overall-survival. Low RagC-expression in the infiltrating zone was as well associated with a longer overall-survival. VPS34 in tumor-center and infiltrating zone and RagC in the tumor-center showed no influence on overall-survival.

Conclusion: Our data suggest that there is a difference in the expression of amino-acid sensors between the tumor-center and the infiltrating zone. MAP4K3 may serve as prognostic marker for the overall-survival. Further investigation on a larger collective is needed to figure out if it is suitable.
