Embocept S® and DC Bead® are more effective to inhibit tumor growth of colorectal rat liver metastases compared to Lipiodol Ultra Fluid®

Introduction: Transarterial chemoembolization (TACE) is an established method in the treatment of primary and secondary malignancies of the liver. However, to date, there are no comparative studies on the different effects of various embolisates. Thus, in the present study we analyzed the effect of 3 different embolisates on the tumor growth in a rat model of colorectal liver metastases.
Material and methods: Eight days after subcapsular implantation of 5x105 colorectal cancer cells (CC531) in the left liver lobe of WAG / Rij rats, randomized into 4 groups (n=8), underwent a TACE by cannulation of the gastroduodenal artery and received either Embocept S®, DC Bead® or Lipiodol Ultra Fluid®. Animals of the control group received a comparable amount of saline solution. Tumor growth was measured on day 8 and 11 using a three-dimensional 40 MHz ultrasound device. On day 11 tumor and liver tissue were taken for histological and immunohistochemical analyses.
Results: On day 11 animals of the control group showed a tumor growth of ~60% compared to day 8. Application of Lipiodol Ultra Fluid® did not significantly influence the tumor growth (~40%). In contrast, the tumor growth of animals from the Embocept S®- and DC Bead®-group was completely inhibited in the same period of time. Immunohistochemical analysis of the tumor cells showed a significant increase of necrotic areas after application of Embocept S® and DC Bead® compared to Lipiodol Ultra Fluid®. However, the proliferation rate of the tumor cells showed no differences between the groups.
Conclusion: The present study demonstrates that a TACE of Embocept S® and DC Bead® results in a complete inhibition of tumor growth of rat colorectal liver metastases in the given time period. Inhibition of tumor growth seems to be caused by increased tumor cell necrosis. Comparable Effects could not be achieved by application of Lipiodol Ultra Fluid®.
