Introduction: Liposarcomas represent 9-18% of all soft tissue sarcomas. They are subclassified into well differentiated, dedifferentiated, myxoid/round cell and pleomorphic liposarcoma. Approximately 50 -70% of patients with retroperitoneal or intraabdominal sarcoma develop a relapse after surgical therapy, including peritoneal sarcomatosis, an extremely rare site of metastatic disease which is associated with poor prognosis. Establishment of permanent cell lines derived from these tumors can be helpful to understand their biological behaviour and to develop new therapeutic strategies.

Material and methods: Liposarcoma cells from ascites of a female patient with peritoneal sarcomatosis were isolated and cultivated. Morphological characterization was performed by immunohistochemistry and IrECM 3D culture. Tumorigenicity was verified in vitro by Invasion-, Migration-, and Soft Agar Assay and in vivo via a subcutaneous xenograft mouse model. Array CGH identified was performed to identify cytogenetic alterations.

Results: Tumor cells isolated from ascites exhibited a spindle-shaped morphology in culture flasks. This newly established cell line (LIPO-DUE1) has been kept in culture for over 25 passages, with a doubling time of 3.5 days. In 3D cultures LIPO-DUE1 cells are morphologically characterized by the “stellate” type. The cells showed invasive and migratory capacity and formed defined colonies in Soft Agar Assays. Four weeks after subcutaneous injection, mice developed local palpable tumors, whereas no distant metastasis could be detected. CGH analysis identified typical amplifications on chromosome 12q in the primary tumor as well as in the cell line LIPO-DUE1.

Conclusion: Peritoneal sarcomatosis is a rare, but aggressive manifestation of liposarcoma. Here, we describe for the first time the establishment and characterization of a liposarcoma cell line derived from a peritoneal sarcomatosis. Hence, the newly established cell line LIPO-DUE1 is a useful in vitro and in vivo model for the investigation of the biological behaviour and tumorprogression of liposarcomas.
