Magnetic tissue engineering using vocal fold fibroblasts treated with superparamagnetic iron oxide nanoparticles – first results of multicellular 3D structures

Zusammenfassung
Hintergrund: Die Stimme ist das wohl wichtigste Instrument der Kommunikation. Ein Defekt an den Stimmlippen, wie beispielsweise nach einer Tumoroperation, verursacht einen deutlichen Verlust der Lebensqualität. Bisher gibt es noch keine befriedigenden Transplantate im Bereich der Stimmlippen. Unser Ziel ist es mithilfe der Nanotechnologie ein funktionelles Stimmlippentransplantat in einem Kaninchenmodel zu generieren. Superparamagnetische Eisenoxidnanopartikel (SPIONs) werden hier im Rahmen des Magnetic Tissue Engineering verwendet.
Material und Methoden: Kaninchen-Stimmlippenfibroblasten wurden für 24 h mit verschiedenen Konzentrationen an SPIONs (5, 20, 40, 60 und 80 µg/cm2) inkubiert. Um eine magnetische Zellsteuerung in zwei Dimensionen zu erreichen, wurde eine 24-Well-Magnetplatte (0,7 T) unter einer 6-Well-Zellkulturplatte mit beladenen Zellen platziert. Die Zellen durften adhärieren und nach verschiedenen Zeitpunkten (12, 24, 48 und 72 h) wurden mikroskopische Bilder aufgenommen. Die Zellen wurden gewaschen, fixiert und mit Kristallviolett, sowie Berliner Blau gefärbt. Um eine Bildung von dreidimensionalen Zellstrukturen herbeizuführen, wurde der Magnet auf eine Zellkulturplatte mit 96 Wells mit 2×104 Zellen pro Well gesetzt. Die dreidimensionalen Zellstrukturen wurden mikroskopisch evaluiert.
Ergebnisse: Lediglich in den Bereichen der Zellkulturplatte, in denen sich ein Magnet befand, kam es zum Wachstum von Zellen. Hierdurch konnte die magnetische Zellsteuerung von mit SPIONs beladenen Kaninchen-Stimmlippenfibroblasten nachgewiesen werden. Durch Platzierung der Magneten oberhalb der Zellkultur bildeten sich bereits nach 24 h dreidimensionale Strukturen von Kaninchen-Stimmlippenfibroblasten.
Fazit: Die Möglichkeit einer magnetischen Zellsteuerung von mit SPIONs beladenen Kaninchen-Stimmlippenfibroblasten wurde in 2D und 3D gezeigt. Es ist gelungen eine dreidimensionale Zellstruktur aus funktionalisierten Kaninchen-Stimmlippenfibroblasten zu erzeugen. Dies ist ein weiterer Schritt auf dem Weg zum Stimmlippentransplantat mittels Magnetic Tissue Engineering.

Introduction
The voice is the most important instrument of communication. Tissue defects in this area, occurring commonly after tumor surgery, lead to serious aggravation in quality of life. Apart from their severe diseases, the patients are faced with social problems, caused by their inability to communicate. Hitherto, no satisfactory vocal fold transplant or implant exists together with a test system that meets demands for biomechanical investigations. Using nanotechnology we aim for the establishment of a functional vocal fold implant in a rabbit model by magnetic tissue engineering with superparamagnetic iron oxide nanoparticles (SPION).
Material and methods
Rabbit vocal fold fibroblasts were incubated for 24 h with different concentrations of SPIONs (5, 20, 40, 60 and 80 µg/cm2). The possibility of magnetic guidance of SPION loaded cells was tested in 2D by using a 24 well magnet plate (0.7 Tesla) placed below a 6 well tissue plate. Cells were allowed to adhere and microscopic as well as macroscopic pictures were taken after 12, 24, 48 and 72 h. Cells were washed, fixed and stained with crystal violet as well as with Prussian blue to identify SPION uptake. To induce 3D cell formation by magnetic levitation 2×104 cells per 96 well were seeded in cell culture medium and magnets (0.7 Tesla) were quickly placed above the culture plate. 3D cell structures were evaluated microscopically.
Results
Magnetically guidance of cells loaded with SPIONs was demonstrated in 2D with 20, 40 and 80 µg/cm2, with cells only growing in areas where a magnet was present even after 72 h (Figure 1 [Fig. 1]). Establishment of 3D cell structure of vocal fold fibroblasts induced by a magnet was visualized after 24 h whereby, 5 µg/cm2 SPIONs were sufficient for this formation (Figure 2 [Fig. 2]).
Conclusion and discussion
Here, we present continuative results of magnetic tissue engineering for voice rehabilitation. As a proof of principle for magnetic cell guidance SPION loaded vocal fold cells were allowed to “choose” either to grow on the side were a magnet or none was placed. Interestingly, 5 to 20 µg/cm2 are sufficient to induce cell growth solitary at site of the magnet in 2D. Furthermore, magnetic cell guidance as only initiator for 3D cell formation was demonstrated to work with very low amount of SPIONs (5 µg/cm2). Next steps include the isolation of epithelial cell and establishment of 3D co-cultures, as well as the proof of functionality in a flow channel model of the rabbit larynx. Our results are a solid basis for a successful transfer of this technique into humans, in order to provide an individual and personalized vocal fold implant.
