Gliederung

Text

To optimise the conditions for cartilage tissue engineering in the laboratory the influence of different factors on the vitality of primary chondrocytes was tested. Former experiments had shown that coating polyethylene surfaces with extracellular matrix molecules facilitates chondrocyte adhesion.

Taking this background into consideration systematical tests were to be carried out to establish to what extent individual origin, number of passages, incubation times and media additives affect the vitality of chondrocytes on PE surfaces.

To determine the vitality of adherent cells results were obtained via MTT or WST tests and compared with the results of the determination of vital cells by trypan blue method.

The expression of extracellular matrix molecules was examined via immunohistochemical analysis and compared with their expression in native septal cartilage.
