Objectives: The nasal epithelium is continuously exposed to commensal inhabitants but their impact on nasal inflammation is still unknown although these species are frequently detectable in the nasal mucosa of patients suffering from chronic rhinosinusitis. We therefore investigated the frequency of S. epidermidis in tissue biopsies from patient with nasal polyps (nasal polyposis, NP) and those without presence of polyps (CRS). In addition, S. epidermidis supernatants were used to induce IL 8 expression in human nasal epithelial cells (HNECs) and in A549 airway cells.

Methods: Sinus mucosa from 30 CRS/ NP -patients was examined for presence of S. epidermidis applying standard and molecular -procedures. For cell culture, HNECs were isolated from healthy turbinate mucosa. Stimulation of HNECs and A549 cells was performed with supernatants of S. epidermidis strains DSM20044 or ATCC35984. IL 8 -synthesis was quantified by ELISA. Inhibition experiments were performed with prednisolone.

Results: S. epidermidis was frequently detectable in the nasal mucosa of patients with CRS and NP. IL-8 levels induced by staphylococcal supernatants were higher in NECs than in A549 cells. Remarkably, addition of prednisolone did not inhibit IL-8 expression in both cell types.

Conclusion: Overall, our results imply pro-inflammatory potential of S. epidermidis on the nasal epithelium. An increased IL-8 expression after addition of staphylococcal supernatants and prednisolone may reflect a synergistic immune response.
