Extensive alterations in laryngeal cartilage macromolecular constituents concentration and structure were observed in squamous cell laryngeal carcinoma, regarding mainly the extracellurar proteoglycans and catabolic enzymes. We have studied these alterations in mRNA level to examine whether they reflected altered mRNA synthesis.

Specimens from 4 healthy individuals and 8 patients who underwent laryngectomy (cancerous and macroscopically normal tissue) were used. Total mRNA was extracted from the tissues using nucleospin RNA II extraction kit, and subjected to RT-PCR amplification and electrophoretic detection.

The expression of versican, Hyal-1, PH-20 and MT1-MMP seemed to be characteristic to the tumor state, since none expression of these macromolecules was observed in healthy samples, however most of these were expressed in normal parts of laryngeal cartilage of laryngectomised patients. Decorin, MMP-2 & -9 and CD44 were expressed in all samples, but their expression was increased in cancer being related with cancer stage. On the other hand, TIMP-1 & -2 expression was decreased in cancer, however they still expressed highly enough in the normal parts of laryngeal cartilage.

The obtained data demonstrated that tumor progression was closely related to the alteration of the expression of specific extracellular constituents. Versican, Hyal-1, PH-20 and MT1-MMP might be used for diagnostic purposes, since they were not expressed in healthy cartilage, and decorin, MMP-2 & -9 and CD44 might help the classification of tumor.
