Expression, activity und localisation of matrixmetalloproteinases in juvenile angiofibroma

Juvenile angiofibroma is a rare tumour almost exclusively occuring in adolescent males. It is a benign neoplasm, which shows an aggressive growth pattern with invasion of adjacent areas. Invasive tumour progression essentially depends on the degradation of extracellullar matrix. Matrixmetalloproteinases (MMPs) and their inhibitors (TIMPs) are important enzymes for the proteolysis of extracellular matrix proteins.
In this study gelatinases, collagenases and the specific inhibitors in several angiofibromas were investigated, concerning expression, activity and localisation. For this purpose western blot, immunofluorescence, zymogramm, in-situ zymography and oligo-array technology were used. The results show, that MMP-2 and MMP-13 are increased in the tumour compared to normal turbinate tissue, whereas the specific inhibitors (TIMPs) were significantly reduced. This could be shown on mRNA level as well as on protein level. MMP-2 and MMP-13 could be localized in the stroma of the tumour detected by in-situ zymography and immunofluorescence.
These results suggest that the imbalance of MMPs and their specific inhibitors (TIMPs) essentially influences the aggressive growth pattern of juvenile angiofibroma.
