In vitro experiments to analyze the effects of 5-ALA incubation time and ciprofloxacin on the efficacy of photodynamic therapy in human meningioma cells

Objective: 5-ALA based photodynamic therapy in meningioma cell lines showed weak effects as compared to glioma cell lines (Tsai, 1999). Recently, an increased PDT efficacy was shown in a cervix carcinoma cell line by addition of ciprofloxacin (Ohgari, 2011). Other unsolved issues in the PDT of meningiomas are ALA- incubation time and -concentrations. Therefore, we proposed to analyse the effect of ciprofloxacin and different incubation times and concentrations in vitro.
Method: An in vitro model of immortalized human malignant meningioma cells was used. In group 1 different ALA concentrations (CALA= 50, 100 and 200 microg/ml) and incubation times (Tink= 4 and 24 h) were defined. In group 2 the control group received PDT after 4 h (CALA= 12.5, 25, 50, 100 microg/ml; Tink= 4 h)) and the experimental group a PDT after the same schema but with additional ciprofloxacin (CCipro= 25 microg/ml). The PDT was performed with a standard protocol with a diode laser (635 nm, 625 s). The efficacy of PDT was measured by LDH-Assay (cell death) and WST1-Assay (cell survival).
Results: Prolonged incubation times increased cell death for both higher ALA-concentrations (CALA 100 and 200 µg/ml) as compared to shorter incubation times (p<0.05). Ciprofloxacin increased ALA-PDT efficacy as compared to the control group (p<0.05). This effect was especially strong for higher concentrations (CALA 50 and 100 µg/ml).
Conclusions: Prolonged ALA-incubation times and ciprofloxacin increased 5-ALA based PDT efficacy in this in vitro model.
